Live cell analysis

Livecyte is a unique system for live cell analysis, which enables Kinetic Cytometry: the analysis of phenotypic and kinetic behaviour of automatically tracked individual cells and cell populations over hours or days.

This advanced technology generates quantitative data without the need for cell labelling.

Exploit inherent cell contrast

Livecyte exploits the inherent contrast mechanisms that cells possess; refractive index and thickness variations.

The Livecyte employs a unique approach that uses a phase retrieval method known as ptychography (tie-cog-rafee) which generates high contrast, high fidelity images without halos or speckling.

When a light wave travels through a cell it is delayed by an amount related to these properties, often referred to as phase shift, phase delay or optical path length. If measured accurately, a map of phase shifts can be related back to thickness and refractive index, thus enabling direct measurement of the physical properties of a cell.

Single cell & population analysis

Single cells can be viewed within the context of cell populations across a large field of view.

System Benefits

To better understand the impact of drug treatments, genetic modifications or environmental factors, it is important to identify and observe all cells in a mixed population.

Livecyte comes complete with automated cell tracking software (patent pending), which can follow all cells for a complete time-course, even if those cells pass over each other.

Low laser power, 10,000-100,000 times less than that used for fluorescence microscopy, means that cells can be imaged, if desired, for greater than 1 week without suffering photo-toxic shock. This benefits all cell types but especially fragile cells such as stem cells and primary cells.

Ptychography allows objective magnification and field of view to be decoupled resulting in large, continuous, high resolution fields of view without stitching for example a 2.5mm x 2.5mm field of view can be captured with a 20x objective lens. A large field of view ensures that highly motile cells are not “lost” during long time-courses. An additional benefit is that multi-area time-lapse regions can be defined in single or multiple wells.

High contrast, high fidelity images lend themselves to more accurate segmentation and this, together with software which identifies and continuously monitors 10 different cellular characteristics for each cell, results in consistently accurate tracking of all cells within a population Multiple cellular measurements are captured for the whole time-course and available for interrogation. Videos of fully segmented cell populations can be generated automatically.

Cells are perfectly focussed irrespective of focal drift or uneven sample holders.

Normalised Total Dry Mass vs Time

Mean Instantaneous Velocity vs Time

Total Dry Mass indicates the amount of cellular material in a field of view. Normalised Total Dry Mass therefore enables a comparison of proliferation rates between multiple wells.

Mean Instantaneous Velocity is a measure of how active, on average, a population of cells is at a given moment in time.

Fast setup

Livecyte’s workflow is designed to be intuitive for all users to setup, collect and analyse their results easily and efficiently. Interactive tools allow experimental protocols to be optimised, saved and re-used.

Virtual eyepiece

A Virtual Eyepiece allows easy navigation and selection of regions of interest and a selection of objective phase and fluorescence illumination wavelengths.

Time lapse

Set up of time lapse assays is simple: an interactive view of the graphical timeline enables stress-free set up.

Fluorescence & Phase

Livecyte enables the capture of phase-only (label-free) images, fluorescence-only images or a combination of both modalities, which can be useful for example, to study the timing of protein expression (labelled fluorescently) in relation to the cell cycle (label free); or when tracking specific translocation events.

10X, 20X & 40X phase

Livecyte allows maximum flexibility regarding objectives. The system is delivered complete with 4x,10x,20x and 40x lenses but a six position turret allows a choice of additional objectives as desired.
